Yes it is possible and by design.

Make sure the computers and users which you wish to apply the policies
to reside within the proper OU. Policies take a little time to apply so
you may have to force this to happen. 

(i.e. XP -- from a command prompt use the command "gpudate /force", this
may or may not require a reboot)

Pay attention to the GPO settings. A common mistake is to try to apply
computer level settings at an OU that contains only users and vise
versa. There is a wealth of information on this at Microsoft's site.
